Description of Cryo‐electron microscopy
Cryo‐electron microscopy (cryo‐EM) is a structural biological method that is used to determine the 3D structures of biomacromolecules. Its purpose is to observe the structure of the sample in its original state as close as possible. It is especially suitable for samples containing water and is an important characterization tool for studying biological samples. Using the solution of sample in-situ freezing–electron tomography–three-dimensional reconstruction, we can obtain the 3D structure diagram of macromolecular biological samples, which is a great tool for studying protein structure. With the development of technology, the application scope of cryo-EM has been further expanded. In addition to common protein structure research, it can also be used to study special materials that are sensitive to electron beams and heat.
